Diagenode Launches State-of-the-Art Solution for Shearing DNA Up to 100 kb Fragments for Long-Read Sequencing

Diagenode’s new Megaruptor 3 can process 1-8 samples in parallel to a fragment range of 5-100 kb using a completely closed system.

DENVILLE, N.J. and LIÈGE, Belgium — Diagenode, a leading global provider for complete solutions for sample preparation and epigenetics research, has launched its third generation long-fragment DNA shearing innovation, the Megaruptor 3. This new solution builds upon the features of the second generation instrument with a user-friendly interface, automated processing, and the ability to process up to 8 samples simultaneously in a completely safe and closed system.

Prior to long-read sequencing on popular sequencing systems such as those from Oxford Nanopore or Pacific Biosciences, genomic DNA must be sheared to long fragment sizes. Though various methods are available for shearing genomic DNA, few are reliable for generating DNA fragments necessary for assembly of microbial, human, plant and animal genomes de novo.

The Megaruptor uses mechanical shearing to fragment DNA. DNA in solution is pumped through the device’s “hydropore” which has an array of uniform pores, stretching and breaking the DNA strands. DNA molecules are passed several times through the pores to guarantee a minimum and uniform length. Additionally, the hydropore design ensures that DNA will not clog the pores, resulting in consistent and efficient shearing not realized by single orifice devices.
