Empowering AAV Quantification for Gene Therapy Research
Introduction
Adeno-associated virus (AAV) vectors have become a cornerstone of gene therapy, valued for their safety, broad targeting capabilities, and adaptability across therapeutic applications.
As development advances from research to process refinement, reliable quantification of total AAV capsid concentration is key to maintaining consistency and supporting informed decision-making.
Traditional methods such as ELISA can be time-consuming, serotype-specific, and sensitive to sample complexity. The Amperia™ platform offers a streamlined alternative – using a sandwich-format assay with high-affinity antibodies from Thermo Fisher Scientific to quantify multiple AAV serotypes with robust performance across sample types. With sensor-based detection and fast turnaround, Amperia enables consistent results in day-to-day workflows.
Assay format & workflow
Amperia™ uses a sandwich assay format for AAV capsid quantification. The sensor surface presents anti-AAV capture sites, typically formed via streptavidin–biotin interaction. AAV particles in the sample bind to this immobilised capture antibody, followed by the addition of a labelled detection antibody specific to the AAV capsid.
As AAV concentration increases, more detection antibody is retained at the sensor surface, generating a stronger signal. This direct signal response supports reliable quantification across a range of sample types, including clarified lysates and crude supernatants.
Assays are run through the system’s touchscreen-guided workflow, using protocol templates tailored for different sample numbers. With intuitive setup and integrated analysis, Amperia enables reproducible AAV titre measurement without the complexity of traditional immunoassays.
