FragIT™ MicroSpin

FragIT MicroSpin columns come prefilled with FabRICATOR® enzyme immobilized on agarose that allow for easy and quick fragmentation of IgG into F(ab’)2 and Fc fragment. Fragmentation of IgG is typically associated with low yield, heterogeneity of the produced fragments and a cumbersome and time consuming process. All these shortcomings can now be solved using FragIT MicroSpin columns for fragmentation of up to 0.5 mg IgG in 15 minutes. After elution from the FragIT™ column, F(ab’)2 and Fc fragments can be separated with size exclusion chromatography.

FabRICATOR®

Genovis has developed a product FabRICATOR, to rapidly generate precise F(ab’)2 fragments. FabRICATOR is a partially modified and his-tagged IdeS enzyme* that cleaves IgG in a singular defined site below the hinge region leaving an intact F(ab?)2 and two residual Fc fragments. The yield of the reaction is high and the enzyme does not further digest the resulting products.

*IdeS is a highly substrate specific cysteine protease originating from Streptococcus

pyogenes.

deGlycIT™ MicroSpin

When performing mass spectrometry of large proteins, such as IgG molecules, there is often a need to remove the glycans because of poor quality of the data otherwise obtained. deGlycIT MicroSpin columns allow for easy and quick deglycosylation of IgG. It is further possible to analyse the degree of fucosylation, since IgGZERO leaves the fucose intact in case the GlcNAc is fucosylated. The columns come prefilled with IgGZERO® enzyme immobilized on agarose and are designed for deglycosylation of up to 0.5 mg IgG in 15 minutes.

IgGZERO™

Genovis has developed a product IgGZERO that is a partially modified EndoS*. IgGZERO cleaves of the side chain glucans leaving the first core N-acetyl-D-glucosamine, or GlcNAc, attached to Asn-297. IgGZERO

* EndoS, a novel secreted protein from Streptococcus pyogenes with endoglycosidase activity on IgG.
